The pen drive records in memory the signals corresponding to the music converted to digital form. There are no audio signals on a thumb drive and therefore cannot be heard simply by connecting it to an amplifier. The circuit used must be able to read digital signals from a memory and convert them to analog (audio) form and then apply them to an amplifier. This is exactly what a pen drive reader does for playing music.
